MHC-matched intravenous adoptive cell transfers
Description of procedure:
Adoptive Transfer Protocol: A donor mouse is euthanized by UCSF standard procedures, and lymphoid cells from donor mice are prepared as a single cell suspension and adjusted to an appropriate final volume with sterile PBS. Each mouse will receive 0. 1- 0.4 ml of cell suspension. Cell numbers may vary between 1 x 10^4 and \1 x 10^8 cells. Recipient mice are injected i.v. via tail vein or anesthetized and injected retro-orbitally using a small gauge (i.e. 30g) needle. If anesthetized, mice will be monitored continuously until ambulatory.
Mice will be evaluated for general appearance, ability to move normally around cage and reach food and water, ability to eat and drink, grooming habits, skin color, lethargy, aggression (animals will be housed individually if aggressive behavior is shown).

| For all investigators
housing animals with tumor formation, skin lesions, neurological
deficits, or that are in Category E, list the expected
characteristics/clinical presentations and endpoints of
the animal model and the criteria for euthanasia. Note:
The IACUC also requires such lists to be posted in the
respective animal rooms and monitored by the IACUC compliance
staff and LARC, to assure PI adherence to the endpoints
listed. |
